Laboratory Experience
Characterization of proteins:
Enzyme assays using chromogenic substrate (activity measurements of proteinase in reactions with inhibitor under various conditions); ELISA; fluorescence-labeling of proteins and studies of the interaction between serine proteinase and inhibitor in a stopped-flow fluorimeter; analysis of protein-protein interactions using surface plasmon resonance in a Biacore; radioactive-labeling of proteins ([32P]).
Expression and purification of proteins:
Culturing of E.coli-, mammalian- and hybridoma-cells; Protein purification using affinity chromatography and size-exclusion chromatography; site-directed mutagenesis with PCR; DNA cloning; generation of cell lines with a stable expression of a chosen protein (using HEK293 Flp-In cells or selection for neomycin-resistant cells); cloning of genes from human RNA using TOPO cloning; transient transfection of COS-7- and HEK293 cells; Western blots of cell extracts.
Cell-assays:
Adhesion-, migration- and invasion-assays; Fluorescence labeling of mammalian cells and quantification using fluorescent microscopy.
Identification of prognostic markers:
Immunohistochemical staining of human bladder cancer tissue sections; statistical analysis of correlation between protein expression in the tumor and the patient response to chemotherapy.
IT-knowledge
Experienced user of Excel and Sigmaplot, the program SwissPDB viewer to display PDB-files of proteins (3D-structures), Word, Internet Explorer, the database PubMed, the drawing program CorelDRAW, and Adobe Illustrator to generate illustrative figures and Powerpoint for presentation of data.

Native langue: Danish.

I have taught students of molecular biology in the laboratory courses: Biochemistry, Molecular Biology, and Cell Biology from the Institute of Molecular Biology, University of Aarhus.
